Background

 

As the major proteins in the basal lamina of the basement membrane, Laminins are trimeric proteins that contain an α-chain, a β-chain, and a γ-chain, which has five (LAMA1, LAMA2, LAMA3, LAMA4, LAMA5), four (LAMB1, LAMB2, LAMB3, LAMB4), and three (LAMC1, LAMC2, LAMC3) genetic variants, respectively. Fifteen laminin trimers have been identified. Lamins form independent and integral network of the structural scaffolding in various tissues. They are associated with type IV collagen networks via entactin, fibronectin, and perlecan and bind to cell membranes through integrin receptors and other plasma membrane molecules to perform their biological functions normally. Laminins play a key role in cell attachment and differentiation, cell shape and movement, maintenance of tissue phenotype, and promotion of tissue survival.

Laminins have been implicated in a number of pathological conditions or pathogenesis of diseases. Dysfunctional structure of laminin-211 has been found to be the cause of one form of congenital muscular dystrophy. It is reported that abnormal laminin-332 is linked to junctional epidermolysis bullos.
