Background

 

IGF II (Insulin-like growth factor II), also known as multiplication stimulating polypeptide/MSP and somatomedin A, is a secreted small polypeptide belonging to the insulin-like growth factor family. IGF II is a potent mitogenic growth factor. However, the growth-promoting-function of IGF II is limited to embryonic development. In human, IGF II is primarily synthesized by the liver, and circulates at high levels in both fetus and adult, while IGF II levels in rodent drop after the perinatal period, suggesting that IGF II has either a limited or local effect postnatally only in rodent. IGF II is expressed in astrocytes, hepatocytes, osteoblasts, and embryonic striated muscle cells in rodent. Mouse IGF II is composed of a 24 aa signal sequence, a 67 aa mature region, and an 89 aa C-terminal prodomain. Mature mouse IGF II is 91% and 97% aa identical to human and rat IGF II, respectively. It binds to IGFIR, the Insulin receptor (IR) type A and IGF IR/IRA complex, the type 2 IGF receptor (IGF 2R), and IGF binding proteins 1- 6 to activate a series of cellular processes.
