Background
E-Selectin, also known as Endothelial Leukocyte Adhesion Molecule-1, ELAM-1, or CD62E, is a type I transmembrane glycoprotein encoded by the SELE gene in humans. E-selectin is composed of an N-terminal, C-type lectin domain, an EGF (epidermal-growth-factor)-like domain, 6 Sushi domain (SCR repeat) units, a transmembrane domain and an intracellular cytoplasmic tail. The rat E-Selectin shares approximately 67% and 78% amino acid sequence identity with human and mouse counterparts, respectively. E-Selectin is expressed only on endothelial cells activated by inflammatory cytokines (IL-1β or TNF-α) or endotoxins.
E-Selectin binds sialyl Lewis X (sLex), a sialic acid-galactose-Nacetylglucosamine-fructose tetra-saccharide. It is reported that this binding is for a specific presentation of those glycosyl units in a precise three-dimensional configuration on a specific glycoprotein rather than for that particular carbohydrate. Cell-surface E-Selectin mediates the rolling attachment of leukocytes to the endothelium, an essential step in extravasation of leukocytes at the site of inflammation, thereby playing a key role in localized inflammatory response. Soluble E-Selectin (sE-Selectin) has been found in the blood of healthy individuals. Elevated levels of sE-Selectin in serum have been reported in a variety of pathological conditions.
