| Applications | NCSTN antibody has been tested by ELISA and Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:500 ~ 1000. Recommended starting dilution is 1:500. |
| Protein Background | Nicastrin is a Type I transmembrane glycoprotein which is an integral component of the multimeric gamma-secretase complex. Nicastrin cleaves integral membrane proteins, including Notch receptors and beta-amyloid precursor protein, and may be a stabilizing cofactor required for gamma-secretase complex assembly. The cleavage of beta-amyloid precursor protein yields amyloid beta peptide which is the main component of the neuritic plaque and the hallmark lesion in the brains of patients with Alzheimer''s disease; however, the nature of the Nicastrin''s role in Alzheimer''s disease is not known for certain. |
