Background

 

The tissue inhibitor of metalloproteinase 3 (TIMP-3), also known as protein MIG-5, is a member of the tissue inhibitor of metalloproteinase (TIMP) family in which four proteins TIMP-1, TIMP-2, TIMP-3 and TIMP-4 have been identified so far. TIMPs are natural inhibitors of the matrix metalloproteinases (MMPs) which are involved in degradation of the extracellular matrix and exhibit diverse functions in development, tissue remodeling, wound healing and tumor metastasis. TIMPs block access of substrates to the MMP catalytic site by forming 1:1 non-covalent complexes with MMPs. Basically, TIMPs are highly specific for MMPs but may not be able to digest some particular MMP such as MMP14. TIMP-3 is expressed by many different cells and is the only known member of the TIMP family that exhibits inhibitory activity on ADAMs such as TACE. It is reported that point mutations in the C-terminal domain of TIMP-3 are correlated with Sorsby's fundus dystrophy.
