Background

 

Myeloid differentiation primary response protein (88) (MYD88) is an adaptor protein encoded by the MYD88 gene in humans. MYD88 is synthesized as a 296 amino acid protein characteristic of a death domain and a TIR domain. MYD88 plays a role in the Toll-like receptor (TLR) and IL-1 receptor signaling pathways in the innate immune response. MyD88 recruits IRAK4 to Toll-like receptors 2 and 4 (TLR2 and TLR4) and plays an important role in the inflammatory response induced by IL-1 and IL-18 and endotoxin. MYD88 can initiate NF-kappa-B activation, cytokine secretion and the inflammatory response by interacting with IRAK1, IRAK2, IRF7 and TRAF6. MYD88 is also involved in IL-18- mediated signaling pathway by increasing IL-18 transcription. MyD88-mediated signaling in intestinal epithelial cells is crucial for maintenance of gut homeostasis and controls the expression of the antimicrobial lectin REG3G in the small intestine. Polymorphisms (SNPs) of the MyD88 gene have been associated with susceptibility to various infective diseases and some autoimmune diseases like ulcerative colitis. It is reported that mutation in MYD88 (L265P) has been identified in many human lymphomas including ABC subtype of Diffuse Large B-cell Lymphoma and Waldenstrom's Macroglobulinemia.
