Hepatocyte growth factor activator, also known as HGF activator, HGFA, is a serum-derived serine protease which belongs to the peptidase family S1. HGFA is first synthesized as an inactive single-chain precursor before being activated to a heterodimeric form by endoproteolytic processing. HGF activator (HGFA) is a serine protease that cleaves the single-chain HGF precursor, generating the active heterodimer. HGFA is secreted by the liver and normally circulates in the blood as an inactive zymogen, which can be activated by thrombin. The active protease is regulated by HGFA inhibitors.
