Background

 

The tissue inhibitor of metalloproteinase 1 (TIMP1), also known as erythroid-potentiating activity (EPA), Fibroblast collagenase inhibitor (Collagenase inhibitor), is a member of the tissue inhibitor of metalloproteinase (TIMP) family in which four proteins TIMP1, TIMP2, TIMP3 and TIMP4 have been identified so far. TIMPs are natural inhibitors of the matrix metalloproteinases (MMPs) which are involved in degradation of the extracellular matrix and exhibit diverse functions in development, tissue remodeling, wound healing and tumor metastasis. TIMPs block access of substrates to the MMP catalytic site by forming 1:1 non-covalent complexes with MMPs. Basically, TIMPs are highly specific for MMPs but may not be able to digest some particular MMP such as MMP14. TIMP-1 is widely expressed by many cells and tissues, and is inducible in response to inflammatory stimuli of IL-1, IL-6, OSM, LIF, TNF-α, TGF-β1 and phorbol esters. In addition to its inhibitory activity on MMPs, TIMP1 also functions as a growth factor that regulates cell differentiation, migration and death and shows erythroid potentiating activity.
