Background

 

The tissue inhibitor of metalloproteinase 4 (TIMP-4), also known as metalloproteinase inhibitor 4, is a member of the tissue inhibitor of metalloproteinase (TIMP) family in which four proteins TIMP1, TIMP2, TIMP3 and TIMP4 have been identified so far. TIMPs are natural inhibitors of the matrix metalloproteinases (MMPs) which are involved in degradation of the extracellular matrix and exhibit diverse functions in development, tissue remodeling, wound healing and tumor metastasis. TIMPs block access of substrates to the MMP catalytic site by forming 1:1 non-covalent complexes with MMPs. Basically, TIMPs are highly specific for MMPs but may not be able to digest some particular MMP such as MMP14. TIMP-4 mRNA is expressed in heart, kidney, pancreas, colon and testes with a higher level at heart. It has been observed that TIMP-4 expression is variable in different cancer types including breast, cervical, and endometrial cancer, gliomas, and choriocarcinoma.
