Background
Heat shock protein of 47 kDa (HSP47), also knwon as serpin H1, clade H, or collagen binding protein 1 is a member of the serpin family of serine proteinase inhibitors. HSP47 is a 418 amino acid molecular chaperone localized at the endoplasmic reticulum lumen (ER) and involved in the collagen folding and secretion. It is shown that HSP47 interacts with collagens I, II, III, IV and V. Its expression is induced by heat shock. Elevated levels of HSP47 and/or autoantibody have been observed in the sera of mixed connective tissue disease patients and other rheumatic autoimmune diseases. HSP47 is also over-expressed in many fibrotic diseases such as systemic sclerosis, pulmonary fibrosis, liver cirrhosis, cicatricial pemphigoid, epidermolysis bullosa acquisita, and keloids. HSP47 might be a potential biomarker or therapeutic target for the treatment of collagen-related fibrosis.
