SerpinA1, also known as Alpha-1 antitrypsin (A1AT), is a prototype member of the Serpin superfamily of the serine protease inhibitors. SerpinA1 is synthesized in hepatocytes and secreted as the abundant proteinase inhibitor in the circulation. SerpinA1 primary target is elastase, but it also has a moderate affinity for plasmin and thrombin. It irreversibly inhibits trypsin, chymotrypsin and plasminogen activator. The major physiological function of SerpinA1 is the protection of the lower respiratory tract against proteolytic destruction by human leukocyte elastase (HLE). SerpinA1 deficiency in circulation is associated with emphysema, liver disease and Alpha-1-antitrypsin deficiency (A1ATD).
