Intended Use
The indirect ELISA kit is designed for detecting the relative level of anti-SARS-CoV-2 (2019-nCoV) Spike RBD or neutralizing antibodies in serum. It can be used for quantifying IgG antibody activity specific for the Receptor Binding Domain [RBD] of the spike protein of the SARS-CoV-2 virus, which causes the COVID-19 respiratory disease, in serum or plasma of vaccinated, immunized and/or infected hosts. This immunoassay is suitable for:
1) Determining immune status relative to non-immune controls.
2) Evaluating efficacy of vaccines, including dosage, adjuvant, route of immunization, and timing.
3) Qualifying and standardizing vaccine batches & protocols. The assay is for research use only and is not intended nor validated for diagnosing SARS-CoV-2 virus disease. Reagents contain no virus or viral antigens.
PRINCIPLE OF THE ASSAY
The wells of Spike RBD IgG ELISA test are coated with Spike RBD recombinant antigen. During the first incubation period Spike RBD IgG or neutralizing IgG if present, will react with Spike RBD antigen bound to the wells. After addition of anti-human IgG HRP conjugate it binds to human IgG to form HRP-human IgG and Spike RBD antigen sandwich. All unbound antibodies and unbound HRP conjugate is washed off. Upon addition TMB substrate, the bound enzyme generates color. The intensity of the color is directly proportional to the concentration of anti-Spike RBD IgG antibodies in the samples.
