Background
CD36, also known as FAT (fatty acid translocase), FAT/CD36, SCARB3, GP88, glycoprotein IV (gpIV), and glycoprotein IIIb (gpIIIb), is an integral membrane protein belonging to the class B scavenger receptor family. CD36, characteristic of two transmembrane segments connected by an extracellular loop and two palmitoylated cytoplasmic tails, is a 472 amino acid (aa) protein containing a 6 aa N-terminal cytoplasmic domain, a 22 aa N- terminal transmembrane segment, a 420 aa extracellular “loopâ€, a 22 aa C-terminal transmembrane segment, and a 9 aa C- terminal cytoplasmic tail. The C- terminal tail of CD36 is involved in oxidized LDL binding. In the extracellular loop, the N-terminal region is shown to bind both thrombospondin 1 and Plasmodium-infected erythrocytes. CD36 can also associate with long-chain fatty acids, collagen, phospholipids and apoptotic cells. The extracellular loop of mouse CD36 is 94% and 84% aa identical to the extracellular loops of rat and human CD36, respectively. CD36 is expressed on capillary endothelium, adipocytes, skeletal muscle cells, intestinal epithelium, smooth muscle cells and hematopoietic cells. CD36 appears to exist as a dimer on cell surface. CD36 plays a role in fatty acid and glucose metabolism. Cooperated with TSP-1, CD36 has shown angiogenic activity. It also participates in the clearance of apoptotic phagocytes. Clinically, CD36 has been implicated in glucose intolerance, atherosclerosis, arterial hypertension, diabetes, cardiomyopathy and Alzheimer's disease.
