Background
Betacellulin (BTC) is a member of the EGF family in which at least ten proteins including EGF, TGFα, amphiregulin, HBEGF, and the various heregulins have been described. All of these cytokines are synthesized as transmembrane precursor proteins and are characteristic of EGF structural units in their extracellular domain. The full-length mouse BTC is a 177 amino acid (aa) residue precursor protein containing a 31 aa signal peptide, an 87 aa residue extracellular region with one EGFlike domain, a 21 aa transmembrane domain and a 38 aa cytoplasmic domain. Mouse BTC shares 93% and 79% aa sequence identity with rat and human BTC, respectively. The soluble forms of these cytokines have been identified and are derived from the membrane bound forms of the EGF proteins by proteolytic cleavage. Human BTC precursor protein exhibits 79% amino acid sequence identity with the mouse BTC counterpart. BTC is a ligand for EGFR, ERBB4 and other EGF receptor family members. It functions as a growth factor and is a potent mitogen for Balb/c 3T3 fibroblasts, retinal pigment epithelial cells and vascular smooth muscle cells.
