Background
Diablo homolog (direct IAP binding protein with low pI), also known as second mitochondria-derived activator of caspase (Smac), is a mitochondrial protein encoded by the DIABLO gene in humans. Several alternatively spliced transcript variants of DIABLO gene have been reported. Diablo is synthesized in mitochondria but is realeased into cytosol when cells undergo apoptosis. Diablo is ubiquitously expressed in various tissues, such as testis, heart, liver, kidney, spleen, prostate and ovary. Diablo homolog has been shown to interact with Lymphotoxin beta receptor, Survivin, XIAP and BIRC2. Primarily, Diablo plays a key role in promoting apoptosis by activating caspases in the cytochrome c/Apaf-1/caspase-9 pathway. Diablo can eliminate the inhibitory activity of inhibitor of apoptosis proteins (IAP) and Inhibits the activity of BIRC6 / bruce by inhibiting its binding to caspases.
