Background

 

Endostatin is a naturally-occurring, 20-kDa C-terminal fragment of type XVIII collagen. Human type XVIII Collagen is composed of an N-terminal region containing at least two splice variants, a central triple helical domain, and a C-terminal, non-collagenous (NC1) domain from which Endostatin is derived. Cathepsin L and Elastase may be responsible for Endostatin generation in EOMA cells. Human monomeric endostatin has a zinc binding domain at the N-terminus of the protein. It binds heparin with high affinity, and binds all heparan sulfate proteoglycans with low affinity. Oligomeric endostatin (trimer or dimer) binds primarily laminin of the basal lamina. Endostatin exhibits potently inhibitory effects on angiogenesis. Endostatin blocks pro-angiogenic gene expression controlled by c-Jun N terminal kinase (JNK) via interfering with TNFα-mediated activation of JNK. It reduces the growth of new cells by inhibiting cyclin D1, leading to cells arrest during G1 phase and enter apoptosis. Endostatin can affect FGF signal transduction and inhibit the migration of endothelial cells through disruption of cell-matrix adhesions, cell-cell adhesions, and cytoskeletal reorganization.
