CD32b, also known as FCGR2B, is a "low affinity" receptor for Immunoglobulin G. The inhibitory CD32b is expressed on B cells and myeloid dendritic cells. CD32b is the inhibitory FcR and the inhibitory capacity of CD32b is largely dependent on expression of an intracellular ITIM. Ligation of CD32b on B cells downregulates antibody production and may promote apoptosis. Co-ligation of CD32b on dendritic cells inhibits maturation and blocks cell activation. CD32b may also be a target for monoclonal antibody therapy for malignancies. Deficiency in CD32b leads to chronic B cell activation and autoimmunity. Polymorphisms in CD32b are associated with an increased prevalence of SLE.
