Background
Insulin-like growth factor-binding protein 6 (IBP-6 or IGFBP-6) is a member of insulin-like growth factor (IGF) binding protein superfamily including the six high affinity IGF binding proteins (IGFBP) and at least four additional low affinity binding proteins referred to as IGFBP related proteins (IGFBPrP). All IGFBP members contain conserved cysteine residues clustered in the N- and C- terminal thirds of the protein. Primarily, IGFBPs modulate the biological activities of IGF proteins. Some IGFBPs may also have intrinsic bioactivity that is independent of their ability to bind IGF proteins. Human IGFBP-6 is synthesized as a 240 amino acid (aa) precursor protein composed of a putative 24 aa residue signal peptide and a O-glycosylated 216 aa mature protein. IGFBP-6 is expressed in ovarian cells, prostatic cells, and fibroblasts. Circulating IGFBP-6 exists predominantly in CSF and serum. IGFBP-6 binds preferentially to IGF-II, exhibiting a 2 fold higher affinity for IGF-II than for IGF-I.
