Background

 

Bone morphogenetic protein-4 (BMP-4), also known as BMP-2b, is a member of the transforming growth factor beta (TGF-β) superfamily. BMPs were originally identified as protein regulators of cartilage and bone formation. Extensive studies have shown that BMP-4 plays an important role in the onset of endochondral bone formation, dorsal/ventral patterning. The BMP-4 precursor is a 408 amino acid (aa) protein containing a 19 aa signal sequence, a 273 aa pro-region, and a 116 aa mature segment. Both the pro-region and mature segment contain two potential N-linked glycosylation sites. The mature region consists of seven highly conserved cysteine aa residues featured in TGF-β superfamily members. The mature region of human BMP-4 shares 98% aa sequence identity with mouse and rat BMP-4. BMP-4 binds to ALK-6/BMP RIB and BMP RII to activate a series of biological processes in which smad proteins play a critical role.
