Background
Tumor necrosis factor ligand superfamily member 12 (TNFSF12), also known as APO3 ligand, TNF-related weak inducer of apoptosis (TWEAK), is a type II transmembrane protein belonging to the TNF superfamily. TNFSF12 is composed of a short cytoplasmic domain, a transmembrane domain and an extracellular domain. TNFSF12 is expressed widely in many different cells and tissues. TNFSF12 is a ligand for Death Receptor 3 (DR3). DR3, also termed TNFRSF12, Apo3, LARD, WSL1, or TRAMP, is a TNF receptor superfamily member that is expressed predominantly in tissues with high lymphocyte content. It is shown that TNFSF12/DR3 induced cell death involves the activation of NFκB. TWEAK receptor (TWEAKR, or FN14) is a novel TNF receptor superfamily member that also binds TNFSF12. FN14 gene is a mitogen-inducible gene that is expressed in fibroblasts, hepetocellular carcinomas and endothelial cells. TNFSF12/FN14 interaction has been shown to play a role in endothelial cell growth and migration. In addition, TNFSF12 has exhibited multiple functions in promoting angiogenesis and the proliferation of endothelial cells, induction of inflammatory cytokines, as well as promoting IL-8 secretion.
