Background

Lyn (Lck/Yes-related novel protein tyrosine kinase), also known as Tyrosine-protein kinase Lyn, V-yes-1 Yamaguchi sarcoma viral related oncogene homolog, p53Lyn, p56Lyn,is a member of the Src family of protein tyrosine kinases. Lyn  is mainly expressed in hematopoietic cells, neural tissues, liver, and adipose tissue. Functioned as a non-receptor tyrosine-protein kinase that transmits signals from cell surface receptors, Lyn plays an important role in the regulation of innate and adaptive immune responses, hematopoiesis, responses to growth factors and cytokines, integrin signaling, as well as  responses to DNA damage and genotoxic agents. Lyn deficient mice display a phenotype that includes splenomegaly, a dramatic increase in numbers of myeloid progenitors and monocyte/macrophage tumors. Mice that expressed a hyperactive Lyn allele were tumor free and displayed no propensity toward hematological malignancy. These mice have reduced numbers of conventional B lymphocytes, down-regulated surface immunoglobulin M and costimulatory molecules, and elevated numbers of B1a B cells.
