Background
The protein group of insulin-like growth factor (IGF) binding proteins consists of six high-affinity IGF binding proteins (IGFBPs) and at least four additional low-affinity binding proteins termed IGFBP related proteins (IGFBPrP). All IGFBP members are cysteine-rich proteins with conserved cysteine residues clustered in the N- and C-terminal thirds of the protein. IGFBPs primarily function as the modulator for the biological activities of IGF proteins, although some IGFBPs may also have intrinsic bioactivity. The 234 amino acid (aa) mature IGFBP 1 protein is derived from the full-length IGFBP 1 precursor by removing the 25 aa N-terminal signal peptide. IGFBP 1 is expressed in liver, decidua, kidneys and is the most abundant IGFBP in amniotic fluid. IGFBP1 binds to IGFI and IGFII with equal affinity, but phosphorylated IGFBP1 has higher binding affinities.
