Background
CCL21, also known as 6Ckine, Beta-chemokine exodus-2, secondary lymphoid-tissue chemokine (SLC), small-inducible cytokine A21, TCA4, Exodus2, and A21, is a small cytokine belonging to the CC chemokine family. Human CCL21 has a 37 amino acid (aa) C-terminal extension to mediate its attachment to carbohydrate structures and extracellular matrix components which is distinct from other CC chemokine members. Mature human CCL21 shares 71% and 66% aa sequence identity with mouse and rat CCL21, respectively.
CCL21 is constitutively expressed on initial lymphatic vessels, high endothelial venules (HEV), and lymph node dendritic cells (DC). Immobilized CCL21 promotes the docking of DC to lymphatic vessels and the retention of T cells by lymph node DC, resulting in T cell priming for activation. Human CCL21 binds to the chemokine receptor CCR7 and transduce its signals in the target cells. CCL2 has been associated with series of pathological conditions. For example, Elevated levels of CCL21 in local vascular endothelial cells, macrophages, T cells, and neurons during chronic inflammation or tissue damage have been observed.
