Background
Human Vascular Cell Adhesion Molecule-1 (VCAM-1), also known as INCAM-100, or CD106, is a type I transmembrane glycoprotein characteristic of seven C2-type immunoglobulin (Ig) domains. This 715 amino acid (aa) protein is composed of a 674 aa extracellular region with multiple N-linked glycosylation sites, a 22 aa transmembrane segment and a 19 aa cytoplasmic tail. Human VCAM-1 is approximately 75% aa identical to mouse and rat VCAM-1. VCAM-1 is expressed in many different cell types such as neurons, endothelial cells, smooth muscle cells, fibroblasts and macrophages. Expression of VCAM-1 in endothelial cells is induced in response to stimuli of cytokines, such as TNFa, IL-1, and IL-4. Soluble VCAM-1 (sVCAM-1) has been found in cerebrospinal fluid, blood, and culture supernatants. VCAM-1 plays an important role in regulating the adhesion of lymphocytes, monocytes, eosinophils, and basophils to vascular endothelium. VCAM-1 is a receptor for both VLA-4 and LPAM-1integrins. Interaction between VCAM-1 and its ligands is crucial to leukocyte extravasation.
