Background
CD105, also known as Endoglin, is a large type I transmembrane glycoprotein acting as a co-receptor for members of the TGF-β superfamily. CD105 contains a disulfide-linked extracellular region, and a short, constitutively phosphorylated cytoplasmic tail. Two alternatively spliced variants (S and L) of CD105 have been identified. The CD105-L form is a 633 amino acids (aa) polypeptide with a 47 aa cytoplasmic region, while CD105-S consists of 600 aa and has a 14 aa cytoplasmic tail. CD105 is highly expressed on vascular endothelial cells, chondrocytes, and syncytiotrophoblasts of term placenta.
CD105 can bind TGF-β1, TGF-β3, BMP-2, BMP-7, and Activin A. CD105 binds TGF-β ligands together with a corresponding receptor to activate the downstream signaling pathways. Although CD105 does not bind TGF-β by itself, it can bind the receptor in the absence of ligand. CD105 may have both positive and negative effects as a modulator of TGF-β signaling. For instance, when CD105 expression is suppressed in HUVEC cells, TGF-β1 effects on proliferation and migration are markedly enhanced, whereas overexpression of CD105 in myelomonocytic cells suppresses TGF-β1 activities. It is shown that CD105 plays a role in hematopoiesis, angiogenesis, and cardiovascular development. CD105 has been used as a marker to define long term repopulating hematopoietic stem cells. It is reported that CD105 knock-out mice display abnormal cardiac development including enlarged hearts, necrosis, and defects in valve and septum formation. In addition, enhanced endothelial expression of CD105 has been associated with angiogenesis in several types of cancer.
