2. One 96-well plate precoated with anti-human CST2 Ab
3. Sample diluent buffer: 12 mL x 1
4. HRP conjugated anti-Human CST2 Ab: 1 vial, dilution 1:500.
5. Detection Antibody diluent buffer: 12 mL x 1
6. 20 x Wash Solution: 25 ml x 1.
7. TMB color developing agent A: 6 ml.
8. TMB color developing agent B: 6 ml.

Background
Cystatin-SA, also known as Cystatin-2, Cystatin-S5 and CST2, is a secreted protein which belongs to thecystatin family. The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins (CST2) and the kininogens. CST2 is expressed in submandibular and sublingual saliva but not in parotid saliva (at protein level). It is also expressed in submandibular gland and parotid gland. CST2 is Thiol protease inhibitor and provides protective functions. Diseases associated with CST2 include autoimmune polyendocrine syndrome type 1.
