Background
L-Selectin, also known as leukocyte selectin, Leukocyte adhesion molecule 1 (LAM-1), Leukocyte surface antigen Leu-8, Leukocyte-endothelial cell adhesion molecule 1 (LECAM1) CD62L, LAM-1, LECCAM-1, TQ1, Leu-8, DREG, lymph node homing receptor, MEL-14 antigen, is a cell surface glycoprotein belonging to the selectin family. L-Selectin is characterized by an NH2-terminal calcium-dependent lectin domain, an EGF-like domain, two CRP-like domains, a transmembrane sequence and a short cytoplasmic domain. Human and mouse L-Selectin show the same domain structure and are 77% amino acid sequence identical.
L-selectin is an adhesion/homing receptor that plays important roles in lymphocyte-endothelial cell interactions. Three ligands (GlyCAM-1, sgp90/CD34, MAdCAM-1) for L-Selectin have been identified on endothelial cells, all containing O-glycosylated mucin or mucin-like domains. Two forms of soluble L-Selectin (sL-Selectin) with different molecular weight have been identified. The lymphocyte form is about 62 kDa, while the fragment derived from neutrophils is 75-100 kDa. sL-Selectin retains bioactivity, and at high concentrations can inhibit binding of lymphocytes to endothelium.
