Background
Factor H is a 155 kDa glycoprotein composed of 20 SCRs (short consensus repeats), each of which consists of approximately 60 amino acids with four invariant Cys residues. It is secreted by Kupffer cells, hepatocytes, vascular endothelial cells, and platelets, and circulates in the serum at high concentration. Factor H plays a key role in negative regulation of the alternative pathway of complement. It is required for prevention of complement activation on host cells and tissues, especially the kidney. Within SCR1520, human Complement Factor H shares 60% and 63% amino acid sequence identity with mouse and rat Complement Factor H, respectively. Factor H controls the formation and decay of the alternative pathway C3/C5 convertase (decay accelerating activity). It also acts as a cofactor for factor I which proteolytically inactivates C3b when C3b is bound to factor H (cofactor activity).
